皮肤

是人体最大的器官,它具有保护、感觉、分泌、排泄、呼吸等功能。 它通过抵御外界微生物的入侵以及排泄体内废物起到维持机体内环境稳定的作 用。健康皮肤的完整性在维持人体生理动态平衡中起着至关重要的作用。当皮肤受到损伤后,糖尿病或压力性溃疡等许多原因可延迟伤口愈合。创面愈合的本质是重建皮肤表皮基底层的角质形成细胞。 在表皮各类细胞当中,角质形成细胞数量占 80%以上,且表皮基底层的角质形成 细胞可以进行细胞增殖并向外层推移分化形成表皮的分层结构,维持着皮肤的稳态。创面治疗方法的根本都是让表皮重新覆盖上角质形成细胞并重新启动上皮化过程,促进皮肤愈合。传统方法如皮肤移植、诱导皮肤干细胞移植技术 (iPS)、皮肤组织工程、皮肤敷料和人造皮肤等在促进角质形成细胞的生成效果并不理想。因此前沿的技术和方法如:
体细胞重编程技术、构建人源胶原蛋白表达体系 以及智能水凝胶敷料来修复皮肤烧创伤是本课题组重点探索的方向。

皮肤 烧创伤修复

利用基因编辑技术结合模式生物斑马鱼构建人源烧创伤修复因子表达系统,包括DGTM(DNP63A、GRHL2、TFAP2A 、MYC)转录因子和XVII 、III型胶原蛋白,用鱼皮作为皮肤烧伤创面的敷料,促进安全有效的烧创伤愈合。 实时监控伤口感染并按需给药治疗的智能水凝胶可适应精准治疗
